Qiagen PCR FAQ
What file format and layout do I need to upload my data into the PCR Array Data Analysis software?
Your data needs to be arranged so that the Raw C(t) or C(p) value corresponds to the correct well location on the PCR Array. A blank spreadsheet is available at: http://www.sabiosciences.com/pcr/template.xls. If you are starting with a 384 well plate which is in the 4x96 layout, then you will need to deconvolve the data using the :384-Well Format E/G Data Analysis Patch tool located at http://www.sabiosciences.com/pcrarraydataanalysis.php
What does a typical EpiTect Methyl qPCR Assay or Array include?
A typical order for EpiTect Methyl qPCR Arrays or Assays includes the EpiTect Methyl DNA Restriction Kit, EpiTect Methyl qPCR Assays or Arrays, and instrument specific SYBR Green master mix.
Why can't I find the DNA methylation qPCR Assay for my gene of interest?
There are three possible reasons. First, there is no predicted CpG island in the promoter region (defined as 5 kb upstream to 3 kb downstream of the transcription start site) of your gene of interest. Second, part of our primer design criteria is that the amplicon should include both methylation sensitive and dependent enzyme cutting sites. If your CpG island of interest does not contain these, we are unable to design primers for it. Third, the gene symbol you input may not be correct. You will be informed of those cases in the search result page.
Why do I need to identify my real-time instrument model when placing my order for RT² qPCR Primer Assays?
The performance of our RT² qPCR Primer Assays have been tested, and are guaranteed with, our RT² SYBR Green Mastermixes only. Different master mixes have been optimized and are available for different qPCR instrumentation, because each instrument uses a different reference dye to normalize their optics. In order to guarantee that your RT² qPCR Primer Assays will work right the first time in your hands, we need to make sure that you receive the correct RT² SYBR Green Mastermix for your real-time instrument.
